Missing secretory granules, dilated endoplasmic reticulum, and nuclear dislocation in the thyroid gland ofrdw rats with hereditary dwarfism

Abstract: Previous studies on the rdw rat have suggested that its dwarfism is caused primarily by dysfunction of the thyroid gland. In this study, rat thyroid glands were analyzed endocrinologically and morphologically to clarify the primary cause of dwarfism in the rdw rat. The rdw rat showed lowered thyroid hormone (T4 and T3) levels but elevated TSH in serum. The rdw thyroid gland was almost proportional in size and it was not goiter in gross inspection. Our histological investigation produced three results that may … Show more

“…The vacuolar degeneration in the present case is very similar to the findings in reported rat cases in which the vacuoles reacted negatively to PAS and thyroglobulin 55 , 58 . Dilatation of RER in follicular cells has also been reported in animals with genetic hypothyroidism 56 , 57 , 59 , 60 , 61 , 62 , 63 . Dilatation of the RER might be the process through which thyroid hormone synthesis or secretion was affected.…”

“…The materials of intracellular vacuoles react positively for thyroglobulin in the dwarfs of GALAS rats 3,4 . The cause of vacuolation was thought to be a mutation of the thyroglobulin gene resulting in retention of misfolded thyroglobulin in r-ERs similar to the mechanism described for the rdw rat, which developed dwarfism by genetic hypothyroidism, and huge vacuoles in the thyroid gland 11 . In contrast, the vacuoles of GALAS rats showing normal growth with thyroid dysplasia were not stained for thyroglobulin in the present study and other reported works 2,4 .…”
