Mitochondria as a therapeutic target for common pathologies

Abstract: Although the development of mitochondrial therapies has largely focused on diseases caused by mutations in mitochondrial DNA or in nuclear genes encoding mitochondrial proteins, it has emerged that mitochondrial dysfunction also contributes to the pathology of many common disorders, including neurodegeneration, metabolic disease, heart failure, ischaemia-reperfusion injury and protozoal infections. Mitochondria therefore represent an important drug target for these highly prevalent diseases. Several strategies… Show more

“…91 Instead, improved autophagy-lysosomal functions by enhancing transport of late endosomes ameliorate ALSlike phenotype. 92 Given the mitochondrial dysfunction and autophagy defects in ALS, 93,94 it may be necessary to combine strategies correcting mitochondrial transport with drugs activating autophagy to improve the mitochondrial quality in ALS ( Figure 2C). “…MitoQ is based on the delivery of a potent antioxidant with targeted lipophilic cations that leads to accumulation up to several‐hundred fold in mitochondria. It has been extensively studied and demonstrated safety in humans . Because increased oxidative stress and subsequent mitochondrial damage are the key mechanisms causing pathogenesis in viral hepatitis, treatment with MitoQ can decrease liver damage in HCV patients .…”

“…It is well known that HSPs expression relies on energy supplying of adenosine triphosphate (ATP), which originates from mitochondria. Mitochondria play many key roles in cell, like oxidative phosphorylation, central carbon metabolism, and the biosynthesis of intermediates for cell growth 20. It was reported that mitochondria‐encoded genes contribute to the evolution of heat tolerance in eukaryocyte 19.…”
