Mitochondrial DNA Copy Number in Peripheral Blood Is Reduced in Type 2 Diabetes Patients with Polyneuropathy and Associated with a MIR499A Gene Polymorphism

Abstract: Our aim was to evaluate in a cohort of 125 Italian patients with type 2 diabetes (T2D), who underwent neurological evaluation, the possible differences in the number of mitochondrial DNA copies (mtDNA) comparing positive and negative patients for cardiovascular autonomic neuropathy (CAN) or diabetic peripheral neuropathy (DPN) and comparing them with healthy controls. We also investigated a possible correlation of the number of mtDNA copies with the polymorphism rs3746444 of the MIR499A. T2D patients show a de… Show more

“…All the groups had similar number of T2D patients and healthy individuals. MtDNA copy number was also found to be decreased in Bahrain and Italian populations ( Al-Kafaji et al., 2018 ; Latini et al., 2020 ). A cohort study in Korean population also reported decreased mtDNA copy number ( Lee et al., 1998 ; Song et al., 2001 ).…”

“…MtDNA copy number, indirect marker of mitochondrial dysfunction ( Malik and Czajka, 2013 ), alterations have been found in several diseases ( Filograna et al, 2020 ). Reduced mtDNA copy number in patients with type 2 diabetes is now well evident ( Al-Kafaji et al, 2018 ; Fazzini et al, 2021 ; Latini et al, 2020 ; Xu et al, 2012 ). Busnelli et al (2019) ) demonstrated indirect relation between reduced mtDNA copy number and oxidative stress along with inflammation.…”

“…It was shown that the homozygous variant genotype for the rs3746444 MIR499A polymorphism correlates with a decrease in the number of mtDNA copies, especially in patients with T2DM ( p = 0.009). The association of the variant MIR499A allele with the mtDNA copy number suggests a possible effect of this polymorphism on oxidative stress ( Latini et al, 2020 ).…”

“…Since NETs represented the target parameter of the primary study, these conditions might have biased the study results, and were therefore matched in the control and septic shock group. Since these conditions are also connected to an impaired mitochondrial function, the matching strategy is also appropriate for this secondary analysis [ 21 , 22 , 23 , 24 ]. Visceral surgical patients underwent major abdominal surgery (MAS), such as Whipple’s procedure, oncological gastric or esophageal resection, or colectomy, whereas all cardiac surgical patients received coronary artery bypass graft (CABG) surgery.…”
