Mitochondriopathies as a Clue to Systemic Disorders—Analytical Tools and Mitigating Measures in Context of Predictive, Preventive, and Personalized (3P) Medicine

Abstract: The mitochondrial respiratory chain is the main site of reactive oxygen species (ROS) production in the cell. Although mitochondria possess a powerful antioxidant system, an excess of ROS cannot be completely neutralized and cumulative oxidative damage may lead to decreasing mitochondrial efficiency in energy production, as well as an increasing ROS excess, which is known to cause a critical imbalance in antioxidant/oxidant mechanisms and a “vicious circle” in mitochondrial injury. Due to insufficient energy p… Show more

“…Neurodegenerative disorders are also characterized by the gradual accumulation of mtDNA mutations that can potentially decrease ETC and ATP production efficiency and increase ROS production [71]. A higher ROS level could cause further mtDNA mutations in a "vicious circle" that leads to cell death [72]. Moreover, abnormalities of the microtubule-associated protein tau (Tau) were observed in various neurodegenerative disorders, including Alzheimer's disease (AD), Parkinson's disease (PD), and Pick's disease [73].…”

“…The mitochondrial respiratory chain is the main production site of reactive oxygen species (ROS) in the cell. Excessive ROS production may lead to mitochondrial damage and apoptosis [ 100 ]. Yao et al [ 58 ] investigated the mechanism of β-elemene resistance to reverse cisplatin (DDP) resistance in lung adenocarcinoma A549/DDP cells.…”

“…Maintaining thermal comfort is energetically costly [47] and demands well-organized mitochondrial functionality. Mitochondriopathies causing dysfunction of the mitochondrial respiratory chain are characteristic for several pathologic conditions such as cancerous Warburg transformation and neurodegenerative disabilities [48][49][50].…”
