MMP-8 C-799 T, Lys460Thr, and Lys87Glu variants are not related to risk of cancer

Abstract: BackgroundSeveral studies have focused on the relationship between MMP-8 variants and cancer risk, but they have been unsuccessful in drawing reliable conclusions.MethodsWe employed odds ratio (OR) together with 95% confidence interval (CI) to assess the correlation between MMP-8 C-799 T, Lys460Thr, and Lys87Glu polymorphisms and cancer risk. We further employed in silico tools to evaluate the effect of MMP-8 expression on cancer susceptibility and overall survival time.ResultsA total of 8140 patients with mal… Show more

“…Genetic polymorphisms of CXCR2 may influence the function of the protein by regulating gene expression. Several previous studies have investigated the correlation of CXCR2 mutations with risk of cancer [32][33][34][35][36][37][38]. However, the conclusions of these studies were not consistent.…”

“…Five genetic models were adopted to evaluate the overall ORs. For the CXCR2 C1208T polymorphism, the five models were allelic contrast (T allele vs. C allele), heterozygous comparison (TC vs. CC), homozygous model (TT vs. CC), dominant (TT+TC vs. CC), and recessive (TT vs. TC+CC) model [37,38]. A Q statistic test was used to calculate the heterogeneity among the included studies.…”

“…Further, this SNP was described to have a reduced risk for basal cell carcinoma (BCC) while exhibited no effect in squamous cell carcinoma (SCC) and melanoma ( 31 ). Conversely, a recent meta-analysis of several MMP8 polymorphisms showed that the K460T (rs35866072) and K87E (rs1940475) variants were not significantly associated with cancer susceptibility ( 32 ). Nevertheless, this SNP was not substantially studied despite its higher prevalence in PTCs of the Western population (80.6%) ( 8 ) and DTCs in our study (86%) suggesting that a future study with a large number of samples is warranted to discover the role of this SNP in thyroid cancer.…”

“…Matrix metalloproteinase 8 ( MMP8 ) is largely secreted by neutrophils and malignant cells [ 9 ]. To date, the function of MMP8 in cancer progression has been confusing as various types of cancers demonstrated different prognosis outcome, despite presenting a similarly high level of MMP8 expression.…”
