Molecular characterization of potato (Solanum tuberosum L.) genotypes using random amplified polymorphic DNA (RAPD) and inter simple sequence repeat (ISSR) markers

Abstract: 19 random amplified polymorphic DNA (RAPD) and five Inter-simple sequence repeat (ISSR) primers were used to characterize 35 potato accessions originating from Mexico, Europe and U.S.A, with the aim of generating information on the genetic diversity and relationship among the genotypes for better exploitation in breeding programs. A total of 222 and 82 loci were generated by the RAPD and ISSR primers, respectively. Both RAPD and ISSR primers revealed 81.45 and 82.98% polymorphism, respectively. Mantel test sho… Show more

“…A total of 55 alleles were polymorphic or 80.9% of the total allele. These results did not differ significantly from the results of Onamu et al (2016) which characterized 35 accessions of potatoes with 19 RAPD primers. However, this result was still lower than Yasmin et al (2006) study which analyzed 6 potato cultivars using 3 RAPD primers with PLP value of 94.29%.…”

“…Molecular markers have been used to explore the population structure and genetic diversity of various crops. Peculiarly in potato, the marker-assisted selection (Ghislain et al, 1999), high-resolution mapping (Meksem et al, 1995), fingerprinting for intellectual property rights claims (McGregor et al, 2000; Duan et al, 2018) genetic diversity (Hoque et al, 2013; Siddappa et al, 2014; Onamu et al, 2016) and phylogenetic studies (Spooner et al, 2005a) were based on AFLP, RFLP, RAPD, and SSR markers. Among molecular marker types, in present research, SSRs were often selected for their high polymorphism level and reproducibility.…”

“…Here, the percentage of polymorphic loci was ranged from 33.33% (by primer OPA-18) to 100% (by primer OPF-08 and OPW-01). Onamu et al (2016). In contrast, a lower level of average polymorphism (~53) was found by Salem and Hassanein (2017) in three potato genotypes with 11 RAPD primers.…”
