Molecular Characterization of Sorghum Chlorotic Spot Virus, a Proposed Furovirus

Abstract: SUMMARYA virus morphologically and physicochemicaUy similar to the type member of the furovirus group, wheat soil-borne mosaic virus (WSBMV), has been isolated from Sorghum bicolor and partially characterized. The virus, sorghum chlorotic spot virus (SCSV), causes symptoms which include distinct elongated chlorotic spots and ring spots as well as yellowing on systemically infected leaves of sorghum and inbred Zea mays lines. SCSV is mechanically transmissible to and produces symptoms on the inoculated leaves o… Show more

“…BSBMV RNAs were extracted from virions in purified preparations with chloroform and phenol (1:1) in 2× STE (34) with 1% sodium dodecyl sulfate (SDS), precipitated in three volumes of ethanol at -20°C and electrophoresed through a 1% agarose-formaldehyde denaturing gel in the presence of 0.5 µg/ml ethidium bromide (10,17,34). Nucleic acid sizes were estimated relative to a 0.24-9.5 kb RNA ladder (Gibco BRL, Gaithersburg, MD).…”

“…Members of this genus typically are composed of two to three particles of varying lengths and lack a 3ṕ oly-A tail (4, 16,17). The question of whether BNYVV should be included in the genus Furovirus because of its distinct qualities has been previously discussed (30).…”

“…P. graminis is economically important because of its ability to transmit a number of different virus species, including soil borne cereal mosaic virus (SBCMV), soil borne wheat mosaic virus (SBWMV), wheat spindle streak mosaic virus (WSSMV), barley mild mosaic virus (BaMMV), barley yellow mosaic virus (BaYMV) (Kanyuka et al 2003;Kühne 2009;Tamada and Kondo 2013), peanut clump virus (PCV) (Thouvenel and Fauquet 1981), sorghum chlorotic spot virus (Kendall et al 1988) and rice stripe necrosis virus (Lozano and Morales 2009). Ward et al (1994Ward et al ( , 1998Ward et al ( and 2005 have described different P. graminis ribotypes based on sequence differences in the ribosomal rDNA region.…”
