Molecular markers of breast axillary lymph node metastasis

Abstract: In breast cancer, axillary lymph node status is one of the most important prognostic variables and a crucial component to the staging system. Several clinico-histopathological parameters are considered to be strong predictors of metastasis; however, they fail to accurately classify breast tumors according to their clinical behavior and to predict which patients will have disease recurrence. Methods based on genome-wide microarray analyses have been used to identify molecular markers with respect to the develop… Show more

“…4,18,37,38 Several other breast cancer markers have been associated with axillary lymph node metastasis such as CCND1, CD44, COX-2, EGFR, HER2/NEU, HPA, and LYVE-1 (reviewed in Ref. 39). In addition, increased expression of vascular endothelial growth factor C (VEGF-C) or VEGF-A, both of which are able to promote lymphangiogenesis in primary tumors, was correlated with an increased incidence of regional lymph node as well as distant metastases in humans and animals.…”

“…[30][31][32] Different techniques assessing protein expression, mRNA expression, chromosome alterations, and DNA copy number changes are being used to evaluate these markers of lymph node metastasis in primary breast tumors. 39 The advantage of SHG detection of Col1 fibers as a biomarker of lymph node metastasis is that the technique is nondestructive and can be used to interrogate live tissues.…”

“…However, most of these prognostic and predictive models have been developed based on primary tumor tissue, whereas metastases, rather than the primary tumor, determine the clinical outcome of cancer patients. It has been shown that metastases, which may develop several years after occurrence of the primary tumor and after prior systemic therapy in the adjuvant or neoadjuvant setting, can differ greatly from primary tumor tissue in terms of genetic characteristics (7)(8)(9)(10)(11)(12)(13). It is therefore anticipated that molecular characterization of metastases will improve the currently available prognostic and predictive models.…”

“…Among the parameters most significantly correlated with lymph node involvement are histological grade, tumor size and age (9,10). Furthermore, number and proportion of evaluated sentinel lymph node biopsies (SLNBs) have been correlated with metastases in the axillary lymph nodes (11)(12)(13)).…”
