Molecular typing of a novel canine parvovirus type 2a mutant circulating in Italy

Abstract: Canine parvovirus (CPV) is the etiological agent of a severe viral disease of dogs. After its emergence in late 1970s, the CPV original type (CPV-2) was rapidly and totally replaced by three antigenic variants named CPV-2a, CPV-2b and CPV-2c. CPV has an evolutionary rate nearest to those of RNA viruses, with consequences on disease diagnosis and epidemiology. This paper reports the molecular characterization of eight CPV-2a strains collected from dogs in Italy in 2016-2017. Genetic analysis was conducted on a … Show more

“…The presence of CPV DNA was confirmed using a primer pair (Table S2) in a PCR protocol amplifying a 700‐bp fragment of the VP2 gene (Touihri et al, ), using the commercial kit GoTaq ® G2 DNA Polymerase (Promega Italia s.r.l. ), as previously described (Mira, Dowgier, et al, ). Amplicons were checked after electrophoresis on a 3% agarose gel supplemented with ethidium bromide.…”

“…Sequence analyses were carried out using primer pairs described by Pérez et al (), using the commercial kit GoTaq ® G2 DNA Polymerase (Promega Italia s.r.l. ), as previously described (Mira, Dowgier, et al, ) with minor modifications (Mira, Canuti, et al, ). After electrophoresis on agarose gel, amplicons were purified and submitted for direct Sanger sequencing with a set of primers, as previously described (Mira, Purpari, Lorusso, et al, ).…”

“…Previous studies provided information on the CPV strains spreading in Italy (Decaro, Desario, et al, ; Decaro et al, , ; Dei Giudici et al, ; Mira, Dowgier, et al, ; Purpari et al, ; Tucciarone et al, ), suggesting the need of a continuous epidemiological survey to evaluate the CPV circulation and evolution, whereas limited data are available on the spread of novel strains imported from other continents (Mira, Purpari, Lorusso, et al, ). The aim of this study was the detection and molecular analysis of CPV strains displaying genetic features of Asian viruses spreading in southern Italy.…”

“…Canine parvovirus 2 (CPV‐2) is classified in the genus Protoparvovirus within the family Parvoviridae (Hoelzer & Parrish, ; Mira, Dowgier, et al., ). CPV‐2 infection is responsible for large numbers of animal deaths worldwide and is one of the most dangerous infectious diseases in young puppies aged 2–6 months.…”
