All the registered Strobilurin‐containing chemicals and one Azole known for its higher efficacy to control the pathogen were compared in a fungicide trial in 2001 on wheat inoculated with Fusarium culmorum. Two application times (BBCH 39 and BBCH 51) were used for the Strobilurin fungicides, whereas the Azole was applied at BBCH 65. Visual disease assessment, enzyme‐linked immunosorbent assay (ELISA), a mycotoxin test, 1000‐grain weight and yield assessment were used for evaluation, and all the techniques in different ways revealed differences in the efficacy of the chemicals. Whereas visual disease assessment showed the effect of the chemicals on symptom development, ELISA and the mycotoxin test indicated differences in the treatments on grain contamination by the pathogen and its mycotoxin, respectively. Assessment on 1000‐grain weight and yield also showed the impact of the treatments in reducing the effect of the pathogen on grain production. Correlation coefficients calculated for the different parameters were significantly higher, indicating that a strong relationship existed among the various parameters used for evaluation. It was thus assumed that any of the techniques can be used to make general recommendations about the effect of chemicals on plant pathogenic fungi such as F. culmorum.

Proper management of plant diseases requires accurate disease diagnosis and precise identification of the phytopathogens involved. Traditional and conventional methods of disease diagnosis and pathogen detection have several demerits. It takes a lot of time to diagnose a disease and to identify a pathogen accurately by traditional approaches. Moreover, results are not always conclusive because similar symptoms can be caused by different pathogens or physiological conditions and sometimes it is very difficult to distinguish closely related organisms on the basis of their morphological characters only. Thus the traditional methods normally applied for pathogen identification and disease diagnosis are less sensitive and less reliable. The need for rapid and accurate identification and diagnostic methods to identify the organisms that cause plant diseases are essential for the formulation of effective disease control strategies.

A Sensitive ELISA for Pythium ultimum Using Polyclonal and Species-Specific Monoclonal Antibodies

A double-antibody sandwich indirect enzyme-linked immunosorbent assay (ELISA) was developed for the detection and quantification of Pythium ultimum. A polyclonal antibody produced to cell walls of P. ultimum was used as the capture antibody, while a P. ultimum-specific mono-clonal antibody (MAb E5) was used for recognition of the fungus. In the ELISA, culture extracts of 7 isolates of P. ultimum exhibited strong positive reactions, whereas none of the 37 isolates of other Pythium spp. and fungal genera had positive reactions. P. ultimum was detected by ELISA in roots of bean, cabbage, and sugar beet seedlings grown in pathogen-infested soil. ELISA optical density readings for infected bean and sugar beet root samples were highly correlated (r > 0.9) with infection levels determined by culturing the samples on water agar. The correlation between the two methods of testing cabbage roots was low, but all cabbage roots in which P. ultimum was detected by culturing were strongly positive in the ELISA. Samples of roots infected with P. irregulare and those with no Pythium infection did not react in the ELISA. The ELISA was highly sensitive; the fungus was detected in culture extracts diluted 1:5,000,000 and in roots with less than 1 infection per 100 cm root.
