Morroniside and loganin extracted from Cornus officinalis have protective effects on rat mesangial cell proliferation exposed to advanced glycation end products by preventing oxidative stress

Abstract: Advanced glycation end products (AGE) are involved in the alterations of renal mesangial cell (MCs) growth, a feature of early stages of diabetic nephropathy (DN). We postulate that morroniside and loganin, 2 components extracted from Cornus officinalis, may ameliorate the detrimental effects of AGE-induced MCs proliferation by preventing oxidative stress. Rat MCs cultured in AGE milieu were treated with morroniside and loganin. Results showed that morroniside and loganin inhibited AGE-induced MC proliferation… Show more

“…19) Furthermore, morroniside has been proven effective in preventing diabetic complications such as diabetic angiopathy and the early stage of diabetic nephropathy by regulating renal mesangial cell growth through inhibiting oxidative stress. 21) Our previous studies showed that Corni Fructus exerted protective effects against hyperglycemia and its related renal damage in streptozotocin-induced diabetic rats. 8) Moreover, we confirmed that morroniside isolated from Corni Fructus could improve hyperglycemia and diabetic nephropathy through preventing advanced glycation endproduct formation and down-regulating the expression of its receptor.…”

“…Loganin is one of the major components of Fructus Corni (Li et al, 1994) and possesses protective effect against advanced glycation end products (AGE) induced rat mesangial cell proliferation (Xu et al, 2006). On the contrary, ursolic acid has been considered as an insulin sensitizing agent with inhibitory effect on PTP (Zhang et al, 2006), although others reported that it also could induce glucocorticoid receptor (GR) binding (Cha et al, 1998).…”

“…8) In particular, morroniside, one of the most abundant iridoid glycosides in Corni Fructus, has been proven effective in preventing diabetic complications such as diabetic angiopathy and the early stages of diabetic nephropathy by regulating renal mesangial cell growth through inhibiting oxidative stress. 9,10) In addition, it exhibits a neuroprotective effect by providing cytoprotection against H 2 O 2 -toxicity in SH-SY5Y cells. 11) Diabetes mellitus is characterized by excessive glucose production.…”
