A novel separating and detecting method of urinary proteins by non-gel sieving electrophoresis-free flow electrophoresis on the microfluidic chip was proposed in this article. Human serum albumin and human transferrin labeled by fluorescein isothiocyanate were mixed and taken as the synthesized urinary protein samples. Factors affected on separation, such as the rate of electric fields applied in two dimension, the addition in the buffer, the concentration, and pH of buffer, were optimized, that is, 10% (w/v) dextran, 0.5% ethanediamine, 0.2 mol L 21 Tris-boric acid, pH 9.3, the ratio of the applied electric field in two dimensions 1:4.8, respectively. Fluorescein isothiocyanate-human serum albumin and fluorescein isothiocyanate-transferrin were efficiently separated in 2 min, and the resolution of two proteins achieved was 2.2 by the designed microfluidic chip. Then, the urine samples of nephropathy patient were analyzed by the proposed nongel sieving electrophoresis-free flow electrophoresis on the microfluidic chip. The results matched well with those detected by common clinical methods.

Copolymers of poly(vinylpyrrolidone) (PVP) and hydroxyethylcellulose (HEC) were synthesized, with PVP to HEC molar ratios of 3∶1, 2∶1 and 1∶1. The copolymers were tested as separation media in DNA fragment separation analysis by microchip electrophoresis (MCE). Separation efficiency over 3.8×10 5 for 118 bp has been reached by using the bare channels without the additional polymer coating step. Under optimized separation conditions for longer read length DNA sequencing, the separation ability of the copolymers decreased with decreasing (PVP-co-HEC) molar ratio from 3∶1 to 2∶1 and 1∶1. In comparison with (PVP-co-HEC) 1∶1, the copolymer with (PVP-co-HEC) 3∶1 ratio showed high separation efficiency. By using a 20 g•L -1 copolymer with (PVP-co-HEC) 3∶1 ratio, ΦΧ174-HaeШ digest DNA marker was successfully separated within 3 min.

In a novel attempt to comprehend the complexity of life, systems biology has recently emerged as a state-of-the-art approach for biological research in contrast to the reductionist approaches that have been used in molecular cell biology since the 1950s. Because a massive amount of information is required in many systems biology studies of life processes, we have increasingly come to depend on techniques that provide high-throughput omics data. CE and CE coupled to MS have served as powerful analytical tools for providing qualitative and quantitative omics data. Recent systems biology studies have focused strongly on the diagnosis and treatment of diseases. The increasing number of clinical research papers on drug discovery and disease therapies reflects this growing interest among scientists. Since such clinical research reflects one of the ultimate purposes of bioscience, these trends will be sustained for a long time. Thus, this review mainly focuses on the application of CE and CE-MS in diagnosis as well as on the latest CE methods developed. Furthermore, we outline the new challenges that arose in 2008 and later in elucidating the system-level functions of the bioconstituents of living organisms.
