N6-Methyladenosine-regulated LINC00675 suppress the proliferation, migration and invasion of breast cancer cells via inhibiting miR-513b-5p

Abstract: Breast cancer (BC) is the most common cancer among women. LINC00675 and miR-513b-5p has been reported to be abnormally expressed in multiple types of cancers and modulate malignant phenotypes of cancer cells. However, to date, the functional role and underlying regulatory mechanism of LINC00675 and miR-513b-5p in BC remains largely unknown. Here, we found that LINC00675 was significantly downregulated in BC tissues and cell lines. Decrease of LINC00675 expression associated with higher tumor grade, lymphovascu… Show more

“…It has been reported that miR-889-3p, miR-660-5p, miR-513b-5p, and miR-450a are related to the proliferation, invasion, and migration of BC, and high expression of miR-660-5p was closely related to lymph node metastasis, advanced TNM stage, and vascular invasion of BC tumors, which might be a promising target for BC treatment [27][28][29][30]. Chen et al have reported the role of miR-4644 in prediction of therapeutic responses and suggest that it could serve as valuable sources for biomarker detections and optimal chemotherapeutic choices for BC patients [31].…”

“…Specifically, the m6A modification on lncRNA mediated by METTL3 can regulate the lncRNA sponge’s competitive endogenous RNA (ceRNA) network activity ( Figure 2 I). Fan et al reported that METTL3 increased the m6A methylation modification of LINC00675 that acts as a ceRNA for miR-513b-5p and could repress miR-513b-5p expression, this m6A modification did not affect the LINC00675 expression but enhanced the interaction between LINC00675 and miR-513b-5p [ 93 ]. In addition, lncRNA could recruit m6A methyltransferase and participate in the m6A modification of the downstream target gene that regulates BC progression ( Figure 2 J).…”

“…Specifically, inhibition of METTL3 diminished the m6A modification of MALAT1, subsequently downregulated the MALAT1 expression to suppress EMT in BC by sponging miR-26b to reduce the expression of HMGA2 ( Zhao et al, 2021 ). Fan et al demonstrated that LINC00675 was a tumor protective factor, and its low expression was associated with higher tumor grade, lymphovascular invasion, and shorter survival ( Fan and Wang, 2021 ). Furthermore, in vitro studies indicated that LINC00675 inhibited BC progression by suppressing miR-513b-5p in a METTL3-related m6A-dependent manner.…”
