Natriuretic action of angiotensin(1–7)

Abstract: Evidence that angiotensin(1-7) (Ang(1-7)) is biologically active and can be synthesized by the kidney prompted us to examine its actions in the rat, isolated kidney. Ang(1-7) had three major effects producing, (1) a substantial natriuresis and diuresis, (2) an increase in urinary sodium concentration associated with a fall in potassium concentration and (3) an increase in glomerular filtration rate without affecting renal vascular resistance. Thus, Ang(1-7) may participate in the renal effects of the reninangi… Show more

“…Pawloski-Dahm and Fink (19) also observed an antidiuretic effect in normal rats chronically infused with Ang- (1-7). In contrast, a natriuretic and diuretic effect of Ang-(1-7) was observed in other studies (16,17). Using isolated perfused rat kidneys, Dellipizzi et al (16) observed a natriuretic effect associated with an increase of urinary volume following infusion of Ang-(1-7).…”

“…Additional studies in our laboratory have demonstrated that Ang-(1-7) has a potent antidiuretic effect in water-loaded rats (14) and that its plasma concentration is increased by hemorrhage or by maneuvers known to increase plasma osmolality such as water deprivation and salt load (15). A natriuretic effect of Ang-(1-7) in isolated rat kidneys (16) or in denervated rat kidneys in vivo (17) has also been described. Ongoing studies strongly suggest that these biological effects are mediated by specific receptors (10,11).…”

“…However, the renal effects of this heptapeptide are incompletely understood. In isolated, perfused rat kidneys and anesthetized animals [172,173], ANG-(1-7) has been shown to increase sodium and water excretion and the glomerular filtration rate (GFR) without affecting renovascular resistance. Hence, it is also possible that ANG-(1-7) induces diuresis and natriuresis by inhibiting renal tubular Na+/K+-ATPase [174,175].…”
