Neurochemical Investigation on the Effects of a New Diphenylpiperazine Calcium Antagonist, KB-2796, on the Central Dopaminergic System of Rats.

Abstract: -The effects of KB-2796, a new diphenylpiperazine calcium antagonist, on the striatal dopaminergic system of rats were investigated in comparison with var ious calcium antagonists and the dopamine antagonist chlorpromazine. The inhibiting effect of KB-2796 on [3H]spiperone binding to striatal membranes in vitro was weaker than those of chlorpromazine and the other diphenylpiperazine analogues, flunarizine and cinnarizine, and more potent than those of verapamil and nicardipine. Diltiazem and nifedipine were in… Show more

“…However, in Japan, the sales of flunarizine have been discontinued for parkinsonism with antidopaminergic action occurred frequently. In contrast to flunarizine, since antidopaminergic effect of lomerizine is weak [17], we could not find reports the onset of parkinsonism in treatment with lomerizine. The efficacy of lomerizine 6 months after initiating treatment has been found to be 71% [18].…”

“…In addition, flunarizine reduces potassium-evoked increases in intracellular calcium in rat cerebral synaptosomes (Kobayahi et al 1992). Flunarizine has also been reported to cause a transient reduction in tyrosine hydroxylase immunoreactivity in nigrostriatal neurons (Takada et al 1992), and to inhibit [3H]-spiperone binding to striatal membranes in a concentration-dependent manner (Ikegami et al 1992).…”
