Nitric Oxide Releasing Aspirin Protects the Gastric Mucosa against Stress and Promotes Healing of Stress-Induced Gastric Mucosal Damage: Role of Heat Shock Protein 70

Abstract: Background/Aim: Nitric oxide (NO) releasing nonsteroidal anti-inflammatory drugs do not cause gastric mucosal damage, despite inhibition of the cyclooxygenase activity to a similar extent as conventional nonsteroidal anti-inflammatory drugs that induce such damage. We compared the effects of native aspirin (ASA) with those of NO-releasing ASA (NO-ASA) on the development and healing of acute gastric lesions induced by water immersion and restraint stress (WRS) and the mucosal expression of heat shock protein 70… Show more

“…Recently, it has been shown that PIO, a natural ligand for PPAR-␥, exhibits its anti-inflammatory effects by suppression of interleukin-1␤, tumor necrosis factor ␣, COX-2, and inducible nitric-oxide synthase and by up-regulation of Hsp70 in a gastric ulcer rat model (39). In line with these data, Ethridge et al demonstrated an inverse correlation of COX-2 overexpression and Hsp70 induction (40).…”

“…Furthermore, the expression of certain HSP proteins, such as HSP 70 and HSP 62, has been implicated in the adaptation of gastric mucosa to aspirin in rats by facilitating cell migration and proliferation [42]. In another report, HSP 70 mRNA was rapidly upregulated by NO-releasing NSAIDs while attenuating stress-induced gastric lesions, suggesting that activation of HSP expression by NO may play an important role in gastroprotection against stress injury [27]. …”

“…The primers for COX-1, COX-2, cNOS, iNOS and HSP 70 were synthesized by Biometra (Göttingen, Germany). The primer sequences were designed according to the published cDNA sequence for the rat COXes, NOSes and HSP 70 [22, 23, 24, 25, 26, 27]. The expected lengths of COX-1 and COX-2 PCR products were 561 and 201 bp, respectively, while the length of cNOS, iNOS and HSP 70 PCR products were 840, 741 and 590 bp, respectively.…”
