NK cell recognition of hematopoietic cells by SLAM-SAP families

Abstract: The signaling lymphocyte activation molecule (SLAM) family of receptors (SFRs) are ubiquitously expressed on immune cells, and they regulate multiple immune events by recruiting SH2 (Src homology 2) domain-containing SAP family adapters, including SAP and its homologs, Ewing's sarcoma-associated transcript 2 (EAT-2) and EAT-2 related transducer (ERT). In human patients with X-linked lymphoproliferative (XLP) disease, which is caused by SAP mutations, SFRs alternatively bind other inhibitory SH2 domaincontainin… Show more

“… 285 , 286 The SLAM family consists of a number of transmembrane costimulatory receptors, such as CD150/SLAM, CD244/2B4, CD84, CD229/Ly-9, CD319/CRACC, and NTB-A. 287 Thus, SAP can bind via its SH2 domain to the ITSMs of various SLAM families of receptors, and this interaction plays a crucial role in mediating the costimulatory signals necessary for T cell activation. 283 Moreover, SAP also exerts its adaptor role by binding to various SH3 domain-containing proteins, such as FYN, PKCθ, βPix, and NCK1, 288 291 thus recruiting them to the SLAM family of transmembrane receptors.…”

“…NK cells are cytotoxic lymphocytes that are critical for the defense against infections and tumors (8). NK cell activation is governed by the integration of activating and inhibitory signals from cell surface receptors (9,10); NK cells detect cells that are under stress as a result of infection or malignancy and rapidly respond by secreting cytotoxic granules or death receptor ligands.…”

“…T-bet binds to the promoter region of the IFN-γ gene and stimulates the protein expression of IFN-γ. CD150 combined with SLAM can bind to the signal molecule SAP and modulate the function of SLAM in IFN-γ production in hematopoietic cells [47,48]. The freeze-dried water extract of Radix Angelicae Sinensis could significantly decreased the expression of T-bet after 12 h of treatment.…”
