Non-alcoholic Fatty Liver Disease and Liver Fibrosis during Aging

Abstract: Non-alcoholic fatty liver disease (NAFLD) and its progressive form non-alcoholic steatohepatitis (NASH) have emerged as the leading causes of chronic liver disease-related mortality. The prevalence of NAFLD/NASH is expected to increase given the epidemics of obesity and type 2 diabetes mellitus. Older patients are disproportionally affected by NASH and related complications such as progressive fibrosis, cirrhosis and hepatocellular carcinoma; however, they are often ineligible for liver transplantation due to … Show more

“…However, further research is required to determine the exact differences among these populations. Nevertheless, it is important to mention that older adults diagnosed with NAFLD are at a heightened risk of several complications, including steatohepatitis and cancer 19 . Besides an increasing age‐adjusted prevalence rate of NAFLD in the elderly, decreasing age‐adjusted death and DALYs rates were observed.…”

“…Similarly, in the liver, we see a significant de-zonation of STING -/-TRMs. The loss of TRMs in these specified niches is likely to predispose the tissue to increased infiltrates, fibrosis and pathogen susceptibility 36,51,52,72,73 . Moreover, the loss of interaction with stellate cells in STING -/livers may have another outcome, liver fibrosis is a common feature of aging livers and considered pathological 72,[74][75][76][77][78] .…”

“…It also helps in the sustainability of structural configuration and contributes to the recovery of tissue function after injury. For instance, during liver fibrosis, a disorder linked to scraping and scratching of the liver, which reduces its functional ability, aging controls the propagation and expansion of ECM-secreting hepatic stellate cells [71]. This response restricts disease progression following liver impairment.…”
