Non-specific adsorption of serum and cell lysate on 3D biosensor platforms: A comparative study based on SPRi

Abstract: Comparative study for non-specific adsorption of cell lysate and serum on recently developed biosensor surfaces is reported. Different surface chemistries including, polyethylene glycol (PEG), a-cyclodextrin (CD), hydrogel dextran and surface initiated polymerization (SIP) based gold surfaces were taken into account for this study. Various techniques including, surface plasmon resonance imaging (SPRi) and matrix-assisted laser desorption ionization tandem time-of-flight mass spectrometry (MALDIÀTOF/TOF MS) tec… Show more

“…This phenomenon might be attributed to the unavoidable interference caused by the non-specific adsorption of proteins present in serum samples, which adversely affects the performance of the biosensor. 36 We conducted a sample addition recovery experiment to assess the accuracy of our biosensor. Under optimal experimental conditions, we detected three different concentrations of ctDNA (0.1, 1, and 10 pM) in PBS containing 1% human serum.…”

“…Finally, the precipitate was filtered using a Buchner funnel and washed several timed with distilled water and ethanol to remove monomers, oligomers and excess oxidant until the filtrate become almost colorless. At 80° C the precipitate was dried for 24h [63][64][65][66].…”
