Nucleotide Sequences of 5' and 3' Non-coding Regions of Pepper Mild Mottle Virus Strain S RNA

Abstract: SUMMARYThe nucleotide sequences of the 5' and 3' non-coding regions of pepper mild mottle virus strain S (PMMV-S) RNA were determined; they are more like corresponding sequences of tomato mosaic virus (ToMV) RNA than those of any other tobamovirus reported so far. The 5' leader contains a 68 nucleotide guanosine-free sequence which differs in several nucleotides from the corresponding sequences in genomic RNA of tobacco mosaic virus (TMV) and ToMV. The messenger activity of PMMV-S RNA in vitro and the polypept… Show more

“…As with other tobamoviruses, the 5' NCR is devoid of G residues and has a short U (5 nt) track at its 5' end. While G residues appear in the leader sequence of TMGMV [38], they do not occur in other tobamoviruses [1,3,23,28,29,41]. The frequently repeated ACA triplet is found 11 times at the leader sequence of ORSV.…”

“…This may reflect the size difference of the 3' NCRs between ORSV and other tobamoviruses. As in 3' NCRs of other tobamoviruses [3,33], the last 378 nt of ORSV appear to be highly structured. In total, nine pseudoknots and a tRNA-like structures are present at the 3' NCR of ORSV [35].…”

“…The last 180 nt ofPMMV-I RNA may be folded in the same possible structure as that previously proposed for other tobamoviruses [6,19,50,60] which consist of three pseudoknots preceeding the tRNA-like structure (Fig. 5 a).…”

“…The 3' end nucleotide sequences of both PMMV-I and P 11 RNAs were determined by the direct cleavage method [48] on fragments obtained by partial RNase T 1 digestion of the 3' end labelled RNAs as described [6]. DNA sequences were determined by the chemical degradation procedure [38] and the dideoxy chain termination method [54].…”

“…The three nucleotides (ACA) upstream and the three (GCA) downstream to the initiation codon are also well conserved. The 5' non-coding region of CGMMV-SH has 60 nucleotides and is 67 to 68~ similar to that of TMV vulgare, TMV-L and a Spanish isolate of pepper mild mottle virus (PMMV-S; Avila-Rincon et al, 1989) or TMGMV. The first nucleotide of the CGMMV-SH genome was found to be G, as are those of the other three (TMV vulgate, TMV-L and TMGMV).…”
