Numerical Taxonomy of Moderately Halophilic Gram-negative Rods

Abstract: A study was made of 516 randomly selected isolates of moderately halophilic bacteria from solar salterns showing salinities between 8.8 and 40.0% (w/v) total salts, located in S.E. Spain. After purification, many cytological, physiological, biochemical, nutritional and an ti biot ic sensitivity characters were determined for 106 selected saltern isolates and two reference strains. Data were coded and analysed by numerical techniques using the Jaccard coefficient ( S J ) , and clusters of strains were obtained … Show more

“…Gram reaction, motility, colony morphology and pigmentation, catalase, urease and oxidase activities, nitrate reduction, Tween 80 hydrolysis, Voges-Proskauer and methyl red reaction were checked as recommended by Sanchez-Porro et al [23]. Acid production from carbohydrates, carbon and nitrogen sources utilization were performed according to Ventosa et al [24]. Also, the susceptibility of the strains to antibiotics was determined on Muller-Hinton agar containing 10 and 20 % NaCl.…”

“…Proteolytic activity of the cultures was screened qualitatively in a saline medium containing skim milk (10%) plus 10% (w/v) total salts [11]. Formation of clear zone around the colonies over the next 48 h was considered as indication of protease production ( Fig.…”

“…Unless otherwise stated, all morphological examinations, biochemical and tolerance tests were performed as described previously (Santos et al 1989;Nunes et al 1992), in 10% MH medium (Ventosa et al 1982) (http:// www.dsmz.de/media/med593.htm) as follows (%, w/v): MgCl 2 , 0.7; MgSO 4 , 0.96; CaCl 2 , 0.036; KCl, 0.2; NaHCO 3 , 0.006; NaBr, 0.0026; proteose-peptone no. 3, 0.5; yeast extract, 1.0 and glucose, 0.1.…”

“…Results were recorded after 4 h incubation at 37°C. Single carbon source assimilation tests were performed in a basal liquid medium described by Ventosa et al (1982) to which filter-sterilized yeast extract (0.1 g/l), ammonium chloride (0.5 g/l) and the carbon sources (2.0 g/l) were added. Growth of the strains was examined by measuring the turbidity of cultures incubated at 37°C as described previously (Moreira et al 2000).…”
