Obesity: Low-Dose Orlistat Effects on Body Weight of Mildly to Moderately Overweight Individuals: A 16 Week, Double-Blind, Placebo-Controlled Trial

Abstract: The use of orlistat 60 mg by mildly to moderately overweight individuals produced significant weight loss in conjunction with a reduced calorie diet and self-instructional materials. This amount of weight loss was associated with improvements in several weight-related risk factors. Orlistat 60 mg may be a useful adjunct to lifestyle measures and has the potential to contribute significantly to weight and risk factor improvement for overweight individuals.

“…These findings are in accordance with previous research demonstrating the efficacy of orlistat 60 mg in reducing body weight and weight-related co-morbidities (Anderson et al, 2006;Anderson et al, 2007). The magnitude of the effects is similar to those reported previously by others.…”

“…Since the effects of orlistat (120 mg) on weight loss and body composition are well established, we did not employ a placebo or diet only group. The precise contribution of orlistat 60 mg per se cannot therefore be determined with certainty; however, the weight loss results were consistent with what has been observed in other trials of orlistat 60 mg (Anderson et al, 2006;Anderson et al, 2007). Previous studies have…”

“…Previous studies included a 4-to 5-week run-in period consisting of placebo medication and a hypocaloric diet before initiating the intervention; a detailed, reduced-energy diet prescription along with the pharmacologic intervention; and participants were primarily middle-aged women with overweight and obesity (9)(10)(11). Comparing our study with a similarly designed trial (8), the magnitude of reported weight loss was comparable (ϳ5% weight loss in response to 16 weeks of orlistat vs ϳ3% with placebo (PϽ0.001) in moderately overweight participants [body mass index 25 to 28]).…”

“…5,15,45 One 16-week research study of adults who combined orlistat with a low-fat, reduced-calorie diet reported that the subjects taking orlistat lost 1.15 kg more than those taking placebo. 46 Some investigators have reported a BMI lowering of up to 4.09 kg/m 2 for patients taking orlistat. 20 However, some research notes no weight reduction benefit from this medication when given to obese adolescents.…”
