Optimization of Extracellular Acid Protease Production From Aspergillus Niger by Factorial Design

Abstract: The cultural conditions for acid protease production by Aspergillus niger was optimised using factorial design experiments and one factor-at-a time approach. In the production medium casein served as substrate and protease activity was measured in terms of tyrosine yield. The yield was further improved through UV mutation. Tyrosine yield amounted to 29.22 mg / g on casein substrate. Protease from this microbial strain was mesophilic. The enzyme was stable over a wide temperature range (30 to 45 °C) with maximu… Show more

“…AgNPs were synthesized under different physiochemical conditions to find optimum conditions favoring stable AgNPs of the smallest size range. The effect of incubation temperature on fungus growth and size of AgNPs was examined employing various temperature ranges, i.e., 25°C, 35°C, 45°C, and 55°C as reported previously [25]. The effect 2…”

“…The requirement for specific nitrogen supplements differs from organism to organism (Sharma et al 2017). In this study, peptone was chosen for use in the fermentation medium, because, among the nitrogen sources used for the composition of the media, this was considered the best source for the production of protease by Pseudomonas fluorescens (Kalaiarasi and Sunitha 2009), Aspergillus niger (Kalaskar et al 2014), Bacillus safensis (Shakir et al 2019). Species such as Rhizopus oryzae, Aspergillus niger, A. oryzae, A. terreus and Cellulomonas bogoriensis have shown optimal activity at 60 °C (Kumar et al 2005;Siala et al 2009;Castro and Sato 2014;Li et al 2016;Raol et al 2016), which was very close to the optimal temperature for M. speciosa proteolytic activity.…”

“…Free radical is one of the factors that induce the pathogenesis of a wide range of age-related degenerative diseases through the oxidative modification of DNA, proteins and vital molecules (Malathi and Chakraborty, 1991;Cohen, 1973). DPPH, a stable radical-generating reagent, was used in the present study for primary screening of the antiradical activities (Yokozawa et al, 1998;Kalaskar et al, 2014). The main antioxidant properties was mainly given by the peptides, which was hydrolyzed by enzyme and acid.…”
