Orlistat and Antisense-miRNA-Loaded PLGA-PEG Nanoparticles for Enhanced Triple Negative Breast Cancer Therapy

Abstract: We demonstrate that orlistat in combination with antisense-miR-21 or current chemotherapy holds great promise as a novel and versatile treatment agent for TNBC.

“…The results did demonstrate an increased bioavailability of the nanoparticle-loaded-drug, which enhanced the therapeutic effect of the chemotherapeutic agents on both MDA-MB-231 and SkBr3 cells in vitro and increased antiapoptotic synergistic effect of the combined drugs compared with the independent use of the drugs. Same results were observed when combining Orlistat and doxorubicin [3].…”

“…However, it is difficult to make a direct comparison of drug dosage between in vitro and human studies. Recent efforts have been made to increase the bioavailability of orlistat through novel delivery methods such as micellar nanoparticles, which has been shown to increase water solubility of orlistat and increase the drug's antitumor properties in vitro and in mouse models [32,33].…”

“…HA-CS-coated PEI-poly(D,L-lactide-co-glycolide) (PEI-PLGA) NPs conjugated with dox and miR-542-3p both improved targeting and increased the uptake of NPs in triple-negative breast cancer cells [18]. Furthermore, delivery of PLGA-PEG NPs encapsulating antisense-miR-21 and orlistat or orlistat NP in combination with Dox significantly enhanced apoptotic effects in MDA-MB-231 and SKBR-3 triple-negative breast cancer cells [19]. In another study, PLGA- b -PEG NPs were successful in delivering anti-sense miR-21 and miR-10b in triple-negative breast cancer [20].…”
