We investigated the relationship between the persistence of uterine bacterial infections with cytologically determined endometritis and ovarian function in 65 postpartum Holstein cows. Vaginal mucus discharges were collected, and endometrial smear samples (n = 130) were collected for cytological and bacteriological examinations from the cows at weeks 5 and 7 postpartum (pp). Blood samples were collected at weeks 3, 5 and 7 pp to determine plasma progesterone concentrations to monitor ovarian activity. According to the bacteriological examination, cows were classified into four groups. The first group (n = 32; 49%) comprised cows negative for bacteria at weeks 5 and 7 pp. The second group (n = 11; 17%) comprised cows with bacterial infections at week 5 pp but that were clear of infection at week 7 pp. The third group (n = 12; 19%) comprised cows without bacteria at week 5 pp but that acquired an infection by week 7 pp. The fourth group (n = 10; 15%) comprised cows with bacterial infections at weeks 5 and 7 pp (persistence of infection). A positive correlation (P < 0.001) was noted between the severity of cytologically determined endometritis, purulent vaginal discharge and the persistence of infection. Cows with persistent infections had a significantly (P < 0.01) prolonged luteal phase compared with cows without infection. In conclusion, the prevalence of cytologically determined endometritis and prolonged luteal phase were significantly increased in cows with persistent infections.

Materials and Methods: A total of 102 cattle which comprised of open cows >90 days postpartum, and heifers with delayed age (>24 months) at first calving were enrolled in the study. Animals were divided into four groups according to their breeds [24 Friesian Sahiwal (FRS), 31 Bradford (BRF), 15 Brangus (BR) and 32 Kedah Kelantan (KK)]. Blood plasma samples for progesterone (P 4 ) analysis using radioimmunoassay (RIA) were obtained twice weekly for about three months. Concurrently, the ovarian activity was also monitored for the development of dominant follicles, corpora lutea as well as any cystic ovarian condition, using a B-mode ultrasound scanner. Results: The highest incidence of AOC associated with either COD or inactive ovaries occurred in Brangus (BR; 53.3%), and the lowest incidence was observed in Kedah Kelantan (KK) cows (12.5%). The difference in AOC was significant (p<0.05) between KK and the other three breeds. In cows, the incidence of COD was highest in FRS (30%), followed by BRF (13.3%), BR (14.3%), and KK (8.7%). Overall, the incidence of COD was higher in beef heifers than in cows. Prolonged luteal phase due to luteal cysts was the predominant type of AOC in the case of FRS (66.7%) while cessation of cyclicity due to follicular cysts was the primary abnormality observed in BR (75%), and BRF (76.9%). Conclusion: This study showed that AOC and COD were the major causes of reproductive failure in both dairy and beef cattle such that the KK cows were the least affected. Moreover, differentiation among the various types of COD was possible by the combined use of P 4 data and ultrasonograms.

Factor XI deficiency was detected in Holstein cows and mummified foetuses in Japan; however, no report is available about the occurrence of Factor XI deficiency in Holstein semen in Japan. Five hundred cows in twelve dairy farms in Hiroshima Prefecture, Japan were under the study. Genomic DNA was extracted from the cows using a commercial DNA kits and screened to Factor XI mutation. Based on the information of the carrier cows found in the cattle population, four Holstein bulls were analysed for Factor XI mutation. DNA was extracted from bull's semen using phenol chloroform method. Extracted genomic DNA of the bull's semen was typed for Factor XI using specific polymerase chain reaction (PCR) primers. The resultant PCR was sequenced using big dye terminator sequencing method. The pedigree of the bulls was investigated. Furthermore, the inheritance of Factor XI mutation to next generation was estimated. Out of the 500 cows, five were heterozygous to Factor XI. Moreover, out of the four bulls, one was found to carry the mutation of Factor XI; it was also a complex vertebral malformation (CVM) carrier. In DNA sequencing, the insertion mutation of 76 bp of poly-adenine that characterizes the Factor XI deficiency was detected in the carrier bull as well as the carrier cows. Pedigree analysis of the carrier bull revealed that his father and mother ID were 2247419A and 14189172A, respectively, that originated from USA Holstein. Out of six daughter cows born to the carrier bull, one cow (16.6%) inherited Factor XI mutation, while three of them (50.0%) inherited CVM mutation. Autosomal recessive genes that affect cow's reproduction have a particular concern to dairy industry. To our knowledge this is the first report of Factor XI mutation in Holstein semen in Japan.
