Oxidative biodegradation of phosphorothiolates by fungal laccase

Abstract: Organophosphorus (OP) insecticides and nerve agents that contain P-S bond are relatively more resistant to enzymatic hydrolysis. Purified phenol oxidase (laccase) from the white rot fungus Pleurotus ostreatus (Po) together with the mediator 2,2P-azinobis(3-ethylbenzthiazoline-6-sulfonate) (ABTS) displayed complete and rapid oxidative degradation of the nerve agents VX and Russian VX (RVX) and the insecticide analog diisopropyl-Amiton with specific activity: k sp = 2200, 667 and 1833 nmol min 3I mg 3I , respect… Show more

“…Based on product structure analysis, it has been proposed that laccase-oxidized ABTS can abstract an H atom from the lignin model compounds, leading to indirect laccase catalysis upon the oxidation of the compounds (25). To date, other types of mediators, particularly phenoxazines and N-OH compounds, also have been recognized for their mediation function in laccase catalysis (1, 6,17,29).Mediated laccase catalysis has been applied to a wide range of applications, such as pulp delignification (9, 10, 12, 22, 32), textile dye bleaching (31), polycyclic aromatic hydrocarbon degradation (16, 23), pesticide or insecticide degradation (1, 29), and organic synthesis (13,28). For the paper and pulp industry, novel biological or enzymatic bleaching technologies (including mediated laccase catalysis) have attracted increasing attention (9,10,12,14,22,27,32) because of concerns regarding the environmental impact of the chlorine-based oxidants currently being used in delignification or bleaching.…”
