Aim To evaluate the effects of hydrophilic dental resin monomers, triethylene glycol dimethacrylate (TEGDMA) and hydroxyethyl methacrylate (HEMA), on the polarization of a human monocyte cell line (THP‐1). Methodology THP‐1 cells were treated with resin monomers at noncytotoxic concentrations for 48 h and were analysed for CD86 and CD206 expressions using flow cytometry. The cells were stimulated for polarization in the presence of resin monomers (co‐treatment) or after treatment with monomers (pre‐treatment). CD86 and CD206 mRNA in co‐treated cells was evaluated using quantitative real‐time polymerase chain reaction. The release of TNF‐α and TGF‐β by pre‐treated and co‐treated cells was assessed using enzyme‐linked immunosorbent assay. Morphological changes of macrophages during polarization were observed using bright‐field microscopy. One‐way analysis of variance was used for statistical analysis. Results TEGDMA (1 mmol L−1) and HEMA (2 mmol L−1) did not induce CD86 and CD206 expressions in THP‐1 cells but rather inhibited their expressions in the co‐treated cells. The inhibitory effects also appeared at the transcription level. However, the expression of surface markers was not affected by pre‐treatment with resin monomers. The release of TNF‐α and TGF‐β by M1‐ and M2‐stimulated cells, respectively, was suppressed by co‐treatment (P < 0.05). Microscopic studies revealed that co‐treatment with resin monomers suppressed polarization‐associated morphological changes such as cell volume increase. Conclusions TEGDMA and HEMA inhibited macrophage polarization to both M1 and M2 at the transcription level, and the inhibitory effects disappeared upon the removal of resin monomers from the cell culture.

The alterations in microenvironment upon chronic arsenic exposure may contribute to arsenic-induced lung carcinogenesis. Immune cells, such as macrophages, play an important role in mediating the microenvironment in the lungs. Macrophages carry out their functions after activation. There are two activation status for macrophages: classical (M1) or alternative (M2); the latter is associated with tumorigenesis. Our previous work showed that long-term arsenic exposure induces transformation of lung epithelial cells. However, the crosstalk between epithelial cells and macrophages upon arsenic exposure has not been investigated. In this study, using a co-culture system in which human lung epithelial cells are cultured with macrophages, we determined that long-term arsenic exposure polarizes macrophages towards M2 status through ROS generation. Co-culture with epithelial cells further enhanced the polarization of macrophages as well as transformation of epithelial cells, while blocking macrophage M2 polarization decreased the transformation. In addition, macrophage M2 polarization decreased autophagy activity, which may account for increased cell transformation of epithelial cells with co-culture of macrophages.

This study tested the anti-osteoporetic effect of quercetin in RBM-induced bone loss model (RBM). After 14-day supplementation of 13cRA to induce RBM, rats were administered with quercetin (100 mg/kg) or alendronate (40 mg/kg). We analysed changes in body and uterine weight of animals, femoral geometric characteristics, calcium and phosphorus content, bone weight index, bone hystology, bone mineral density (BMD), markers of bone turnover, lipid peroxidation, glutathione levels and SOD, CAT activity of liver, kidney spleen, and ovary as well as biochemical and haematological variables.In comparison to the control RBM rats, the treatment with quercetin increased bone weight index, BMD, osteocalcin level, femoral geometric characteristics, calcium and phosphorus content in the 13cRA-induced bone loss model. Histological results showed its protective action through promotion of bone formation. According to the results, quercetin could be an effective substitution for alendronate in 13cRA-induced osteoporosis.
