Paper ionophoresis of carbohydrates. I. Procedures and results for four electrolytes

Abstract: Details are given of procedures for effecting paper ionophoresis of polyhydroxy compounds in the electrolytes borax, sodium arsenite, basic lead acetate, and sodium hydroxide. and for detecting the compounds after ionophoresis. Rates of migration are reported for 96 compounds, including all pentose and hexose sugars, the common disaccharides, all sugar alcohols up to the heptitols, the cyclitols, a number of glycols, and several glycosides and other derivatives of carbohydrates. Some new or improved reagents h… Show more

“…long, between horizontal glass plates, I -4 cm. thick, in 0.05 Msodium tetraborate (Frahn & Mills, 1959); a potential difference of 800-900 V was applied for 5 hr; cooling was done by doing the experiments in a refrigerator. Carbohydrates were detected with a modified alkaline silver nitrate reagent (Frahn & Mills, 1959).…”

“…2A). After periodate oxidation, reduction and dephosphorylation (12,14,30) iditol was the only alditol detected (11,14). iditol from the InsP3 was resistant to L-iditol dehydrogenase even under conditions where an internal marker of L-iditol was entirely oxidized (29), and this shows that it was D-iditol (Fig.…”

“…(Brown, 1970) and (ii) 0.25 mm layers of cellulose with doubledevelopment in butan-I-ol/pyridine/water (6 : 4 : 3, by vol.). Reducing sugars and oligosaccharides were detected by spraying with aniline phosphate and heating for 20 min at I I 5 "C (Frahn & Mills, 1959). Glucose and cello-oligosaccharides appeared as brown spots but their yellow fluorescence under ultraviolet light [Mineralight (UVSL-58) UJ tra-violet Products, California, U.S.A.] provided a more sensitive method of detection.…”
