Parental competition for the regulators of chromatin dynamics in mouse zygotes

Abstract: The underlying mechanism for parental asymmetric chromatin dynamics is still unclear. To reveal this, we investigate chromatin dynamics in parthenogenetic, androgenic, and several types of male germ cells-fertilized zygotes. Here we illustrate that parental conflicting role mediates the regulation of chromatin dynamics. Sperm reduces chromatin dynamics in both parental pronuclei (PNs). During spermiogenesis, male germ cells acquire this reducing ability and its resistance. On the other hand, oocytes can increa… Show more

“…4 ). Indeed, the H3.3 level in rs-mPN was significantly lower than that in sp-mPN [ 54 ], which is consistent with our findings because the retained histone H3 from round spermatids presumably prohibited zygotic H3 deposition. Thus, in addition to oogenesis, spermiogenesis is important for chromatin structure and associated gene expression in early preimplantation embryos.…”

“…To examine the mechanisms underlying asymmetric parental histone mobility, several types of zygotes prepared using a micromanipulator were examined [ 54 ]. We hypothesized that protamine-histone replacement is important for the acquisition of high histone mobility in male PN.…”

“…A 5-EU assay of the embryos was performed as described previously ( 32 ), with slight modifications. Briefly, the embryos were transferred to KSOM medium containing 2 mM 5-EU (C10330, Thermo Fisher Scientific) and cultured for 2 h. Then the embryos were fixed with 3.7% PFA for 20 min at room temperature.…”
