Parthenogenetic activation of bovine oocytes using single and combined strontium, ionomycin and 6-dimethylaminopurine treatments

Abstract: SummaryIn vitro-matured (IVM) bovine oocytes were activated with single and combined treatments of strontium (S), ionomycin (I) and 6-DMAP (D). Using oocytes IVM for 26 h, we observed that activation altered cell cycle kinetics (faster progression, MIII arrest, or direct transition from MII to pronuclear stage) when compared to in vitro fertilization. ) and 30 h (50.0%). We also observed that IS allowed higher haploid (77.4%) embryonic development, whilst ID was better for diploid (89.1%) development. It was c… Show more

“…The low blastocyst rates obtained when fertilization was performed at 28 h of IVM might be explained by the fact that fertilization does not occur immediately after insemination. According to Méo et al (2007), the pronuclei first appear 12 h after insemination. Thus, oocytes inseminated later (28 h) would have missed the optimal moment for fertilization due to aging.…”

“…The Ca 2+ ionophore ionomycin has also been shown to induce [Ca 2+ ] i elevations in occytes of many species. According to the literature, the parameters of ionomycin treatment used to activate oocytes of various species vary significantly: concentrations ranging from 5 μM to 5 mM [33] and treatment duration of 1 min to 40 min have also been reported [34]. The influence of ionomycin on cat oocytes has never been described; therefore optimizing the concentration and incubation time is critical for successful oocyte activation.…”

“…For the oocyte activation protocol, 5 µM of ionomycin for 5 min was used. Then, oocytes were washed one more time and cultured in 2 mM 6-dimethylaminopurine for 4 h (Méo et al, 2007). Finally, parthenogenetically activated oocytes were cultured in modified synthetic oviduct fluid (SOF) supplemented with 5% FBS, 100 IU/ml of penicillin and 50 μg/ml of streptomycin under 5% CO 2 , 5% O 2 and, 90% N 2 at 38.5°C, up to Days 7, 9 or 12, when the conditioned medium (CM) was stored at -80°C for subsequent usage.…”
