Pharmacokinetics and Bioavailability of the Anti-Human Immunodeficiency Virus Nucleotide Analog 9-[( R )2-(Phosphonomethoxy)Propyl]Adenine (PMPA) in Dogs

Abstract: The pharmacokinetics, bioavailability, and metabolism of the anti-human immunodeficiency virus nucleotide analog 9[(R)-2-(phosphonomethoxy)propyl]adenine (PMPA) were determined in beagle dogs following intravenous, intraperitoneal, and oral administration. Fasted male beagle dogs (n ‫؍‬ 5) were pretreated with pentagastrin and received PMPA (10 mg/kg of body weight) by the intravenous and oral routes with a washout period of 1 week between doses. A further group of male dogs received PMPA as a single dose via … Show more

“…Although adefovir is used to treat Hepatitis B virus infection, it is unlikely to be co-administered with TNF to a patient with both HBV and HIV infection since TNF is also effective against the former infection [10]. Additionally, adefovir was used as an IS for TNF assays by Hazra et al [8], Sparidans et al [6] and Cundy et al [11]. Fig.…”

“…16 Proximal renal tubular dysfunction and Fanconi's syndrome have been reported to be associated with TDF usage. [10][11][12][13][14][15][17][18][19] ␤ 2 -Microglobulin is commonly measured in urine by enzyme-linked immunosorbent assay (ELISA) or latex agglutination assay.…”

“…[5][6][7][8][9][10][11][12][13][15][16][17] Among those, some used a gradient elution program [6][7][8][9]11,13,17 and/or mass spectrometry, [10][11][12][13]16,17 which imposes higher instrument requirements for the laboratory. Others used fluorescent derivatization that required a special reagent that is not commercially available in China.…”

“…Fluorescent derivatization has been used to determine TFV concentration in other studies. 5,9,15 Unfortunately, the reagent used to derivatize TFV (i.e., chloroacetaldehyde) is not commercially available in China due to its high toxicity. Furthermore, the process of derivatization is elaborate and not easy to control.…”
