Pharmacokinetics of Paeoniflorin After Oral Administration of Shao-yao Gan-chao Tang in Mice

Abstract: ABSTRACT-Paeoniflorin, a monoterpene glycoside, is the principal bioactive component of Paeoniae Radix. The traditional prescription Shao-yao Gan-chao Tang (SGT; Kampo: Shakuyaku-Kanzo-To), which is composed of Paeoniae Radix and Glycyrrhizae Radix, has been widely used in China and Japan. Quantification of paeoniflorin in mouse plasma after oral administration of SGT (at a dose containing 10 mg /kg paeoniflorin) was achieved using a simple and rapid high-performance liquid chromatography method. The plasma co… Show more

“…An earlier study indicated that higher plasma concentrations of paeoniflorin were achieved by using a more crude extract of the plant (6). Additional reports showed that higher plasma concentrations of paeoniflorin were achieved when it is included as a part of multiple herb formula: Shao-yao Gan-chao Tang (6,7). These results indicated that the bioavailability of paeoniflorin could be improved by other phytochemicals present in the herb or herbal formulations.…”

“…In the present study, therefore, we investigated the time course of paeoniflorin in rat hippocampus after intravenous administration of Paeoniae Radix extract. Though a few HPLC-UV methods were reported to determine paeoniflorin in crude drugs containing Paeoniae Radix [28,29] and in animals' plasma after oral administration [30,31], little work of paeoniflorin on cerebral nucleus has been carried out due to the lack of methodologies that meet the requirements of sensitivity and specificity for paeoniflorin determination in hippocampus. In the present study, we successfully developed a rapid and simple HPLC method to determine paeoniflorin in rat hippocampus.…”

“…Thus, it is important to quantify albiflorin and paeoniflorin in rat serum and investigate their pharmcokinetics for evaluating the clinical applications of Si-Wu decoction. There are some reported quantitative methods and pharmacokinetic studies on paeoniflorin in biological samples [19][20][21][22][23][24]. But simultaneous determination of albiflorin and paeoniflorin in rat serum and their pharmacokinetic study were not previously reported.…”
