Photostimulation of Phox2b Medullary Neurons Activates Cardiorespiratory Function in Conscious Rats

Abstract: Photostimulation of ChR2-transfected RVLM Phox2b neurons produces a vigorous stimulation of breathing accompanied by a small sympathetically mediated increase in BP. These results demonstrate that breathing can be relatively selectively activated in resting unanesthetized mammals via optogenetic manipulation of RVLM neurons presumed to be central chemoreceptors. This methodology could perhaps be used in the future to enhance respiration in humans.

“…Moreover, active expiration occurs after photoactivation [68] . On the other hand, photoactivation of C1 neurons in the rostral ventrolateral medulla (RVLM) in which ChR2 is expressed increases sympathetic activity and blood pressure [40,69] . These findings provided the direct evidence that C1 neurons play a role in sympathetic-promoting activity and blood pressure regulation.…”

“…Retrotrapezoid nucleus neurons are directly activated by CO 2 /H + , as demonstrated in neuronal recordings from brain slices and acutely dissociated preparations . Retrotrapezoid nucleus neurons are glutamatergic, and their selective stimulation in vivo results in rapid and robust breathing activity Kanbar et al 2010), while selective inhibition blunts whole-animal breathing responses to hypercapnia Takakura et al 2014), thus indicating that RTN chemoreceptors provide an excitatory drive to breathe.…”
