Background: In this study, to investigate whether piperine, an alkaloid from Pipper longum, could potentially exerts its effect for the suppression of inflammation and oxidative stress, we examined the modulatory effects of piperine in tert-butylhydroperoxide (t-BHP)-induced Ac2F rat liver cells. Methods: Anti-inflammatory mechanism of piperine in Ac2F cells were examined by performing western blotting. Results: The piperine exhibited remarkable reduction of intracellular reactive species (RS) levels in t-BHPand SIN-1-induced Ac2F cells. In addition, piperine inhibited t-BHP-induced activation of nuclear factor kappa B (NF-κB) by suppressing the degradation of inhibitor-κB proteins (IκBα) and translocation of p65 from the cytosol to the nucleus, further indicating piperine's inhibitory effects on nitric oxide synthase (iNOS), cyclooxyganse-2 (COX-2) expressions. As a consequence, piperine modulated through inhibition of ERK, JNK, and p38 MAPKs signal transduction pathway in cells. Moreover, piperine pretreatment also regulated the protein expression of antioxidant enzymes such as manganese-dependent superoxide dismutase (MnSOD) and catalase. Conclusion: These results indicated that piperine, a major component of black pepper might be a potential antiinflammatory agent by modulating RS-induced NF-κB activation through the MAPKs signaling pathway and possess the anti-oxidative property. Therefore piperine can be considered as a useful therapeutic and preventive approach for the treatment of inflammation and oxidative stress-related diseases.

Herbal medicines are known to mitigate radical induced cell damage. Hence identification and scientific validation of herbal medicines contribute to better use in Ayurvedic/Unani research. In the present study, we investigated antioxidant and anti-apoptotic properties of Convolvulus pluricaulis (C. pluricaulis). C. pluricaulis exhibited antioxidant potential evident by free radical scavenging activities. C. pluricaulis pretreatment inhibited H2O2 induced macromolecule damage such as plasmid DNA damage and AAPH induced oxidation of bovine serum albumin and lipid peroxidation of rat hepatic tissues. Further to identify the neuroprotective properties of C. pluricaulis, SHSY5Y cells were treated with H2O2 with or without pretreatment of C. pluricaulis. The C. pluricaulis pretreatment at 50 μg/ml dose exhibited 50% cell survival against 100 μM H2O2 challenge for 24 h and it also decreased the lactate dehydrogenase leakage. Further C. pluricaulis pretreatment restored and regulated the antioxidant and apoptosis markers such as SOD, CAT, p53, and caspase-3 and inhibited, reactive oxygen species generation and depolarization of the mitochondrial membrane. C. pluricaulis possess a high content of flavonoids and polyphenols and GC-MS and FTIR analysis showed a wide variety of compounds which may contribute to the observed effects.

Background: Celastrus paniculatus is a herb used in the Ayurvedic system of medicine that has been reported to show multiple pharmacological properties. In this study, we explored the antioxidative, hypolipidaemic and hypoglycaemic potential of C. paniculatus methanolic seed extract (CPMSE) in high-fat diet (HFD)/streptozotocin (STZ)-induced type 2 diabetes mellitus (T2DM) rats.Materials and methods: Seeds of C. paniculatus were extracted in methanol using Soxhlet extraction method. A total of 36 rats were induced with STZ and HFD and treated with glibenclamide or various concentrations of CPMSE. Upon treatment, blood samples were collected and kidney and liver samples were homogenised. Serum biochemical estimation was performed using several diagnostic kits. Protein was estimated by bicinchoninic acid (BCA) method. Oxidative stress was assessed by measuring malondialdehyde level and superoxide dismutase (SOD), catalase (CAT) and glutathione-S-transferase (GST) activity.Results: CPMSE caused improvements in glucose homeostasis, lipid profile, liver function and oxidative stress parameters in a dose-dependent manner. CPMSE significantly decreased the levels of fasting blood glucose and glycated haemoglobin as well as increased insulin level and total protein content. There was an increase in total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C), triglycerides (TG) levels and reduction in high density lipoprotein-cholesterol (HDL-C) level. There was a decrease in serum levels of serum glutamate pyruvate transaminase (SGPT), serum glutamate oxaloacetate transaminase (SGOT) and alkaline phosphatase (ALP). CPMSE decreased LPO and increased CAT, SOD and GST activity.Conclusion: CPMSE has hypoglycaemic, hypolipidaemic and antioxidant properties by reducing the oxidative stress.Keywords: diabetes mellitus, Celastrus paniculatus, antioxidant, phytochemicals, phytonutrients, streptozotocin, high-fat diet
