Pleiotropic regulatory genes bldA , adpA and absB are implicated in production of phosphoglycolipid antibiotic moenomycin

Abstract: Unlike the majority of actinomycete secondary metabolic pathways, the biosynthesis of peptidoglycan glycosyltransferase inhibitor moenomycin in Streptomyces ghanaensis does not involve any cluster-situated regulators (CSRs). This raises questions about the regulatory signals that initiate and sustain moenomycin production. We now show that three pleiotropic regulatory genes for Streptomyces morphogenesis and antibiotic production—bldA, adpA and absB—exert multi-layered control over moenomycin biosynthesis in n… Show more

“…4B). The enhancement of DNR production in bldA-25 strain cannot be credited solely to translation of a TTA codon in dnrO, since bldA also plays important role as pleiotropic regulator (Makitrynskyy et al 2013). However, the bldnrO-25 strain produces 5.69 mg/l of DNR, providing the evidence to visualize the role of bldA in TTA codon translation (Fig.…”

“…The well-known target of bldA-tRNA is adpA regulator Streptomyces species which regulates production of gamma-butyrolactone (A-factor) known for activation of aerial mycelium formation and secondary metabolism (Makitrynskyy et al 2013). While looking at bldA dependence pattern of TTA codon present in Streptomyces genes, nucleotide "C" or "T" followed TTA codon on eight instances out of nine genes tested (Trepanier et al 2002).…”

“…The application of the OSMAC approach (cultivation of one strain under different conditions) to wake up silent gene clusters may sometimes not be enough. Genetic manipulation of regulatory genes, such as the pleiotropic regulator genes adpA 44 and bldA 45,46 , is also an effective method to activate the production of other secondary metabolites.…”

“…This prompts to investigate the mechanisms of regulation of Mm biosynthesis. In this regard, the transcriptional and translational regulatory mechanisms are studied relatively well [13, 14]. Yet, the role of LSMs in Mm biosynthesis remained unstudied.…”
