Polydopamine-Encapsulated Perfluorocarbon for Ultrasound Contrast Imaging and Photothermal Therapy

Abstract: Biomedical nanoplatforms have been widely investigated for ultrasound (US) imaging and cancer therapy. Herein, perfluorocarbon (PFC) is encapsulated into biocompatible polydopamine (PDA) to form a theranostic nanosystem, followed by the modification of polyethylene glycol (PEG) to stabilize the nanoparticle via a facile one-pot method. Under 808 nm near-infrared laser irradiation, PDA can generate hyperthermia to transform PFC droplets to bubbles with high US imaging sensitivity. The US imaging detection of th… Show more

“…In other studies, Shuai and coworkers developed various gas-filled polymeric carriers for cancer treatment [ 29 , 131 , 132 ]. For example, a theranostic nanodroplet with encapsulated perfluoro- n -pentane (PFP), pentafluorobutane (PFB), and DOX (PFP/PFB/DOX-PPEHD) were used for imaging and as an anticancer drug vehicle [ 129 ].…”

“…[187] The incorporation of photothermal agents in PFC droplets allows for optical droplet vaporization by laser light, which could be advantageous for photoacoustic imaging. [190,191] Collagen disruption and ECM remodeling can be induced using external forces as well, by applying pulsed high intensity-focused US. As a result, tumor targeting and penetration of fluorescent chitosan nanoparticles was improved in mice ( Figure 9D).…”

“…These compounds are widespread in consumers life through plastics, fire retardants, dyes, surfactants, polymers, and pharmaceuticals, among others [1][2][3][4][5][6]. Benign PFCs have been used in the development of biomedical applications, such as emulsions, [7,8] imaging agents, [9,10] biocompatible lubricants, [11] oxygen therapeutics, [12] pulmonary delivery agents, [13] and theranostic agents [14]. On the other hand, perfluoroalkyl acids (PFAs) and fluorinated greenhouse gases (F-gases) belong to a class of persistent chemicals, widely used in industrial and commercial products [1,2,5,6].…”
