pPeOp from Omphalia lapidescens Schroet induces cell cycle arrest and inhibits the migration of MC-4 gastric tumor cells

Abstract: The aim of the present study was to investigate the effect of purified Omphalia lapidescens protein (pPeOp) extracted by polyvinylpyrrolidone from the fungus Omphalia lapidescens Schroet on the proliferation and cell cycle progression of MC-4 human gastric tumor cells. Using polyvinylpyrrolidone, pPeOp was extracted from O. lapidescens Schroet. MC-4 cells were cultured with 30, 60 or 90 µg/ml pPeOp, with 5-fluorouracil used as a positive control. Survival rates of treated cells were significantly decreased com… Show more

“…The human normal gastric cell line MC-1 and human gastric cancer cell lines MC-4, which had been reported at previous study [8], were provided from Zhejiang Provincial Center for Disease and Prevention Centre (Zhejiang CDC). Human normal gastric cell line MC-1 and human gastric cancer cells were cultured with RPMI-1640 medium with 5% (v/v) fetal bovine serum (Gibco®), 100 U/ml penicillin and 100 U/ml streptomycin in a humidified 5% CO2 atmosphere at 37°C.…”

“…pPeOp is a protein extracted from O.lapidescens with PVP extraction buffer [7]. Previous studies demonstrated that pPeOp suppresses metastasis, arrests cell cycle and induces apoptosis in MC-4 gastric cancer cells without causing toxicity to MC-1 normal gastric cells [8]. Thus, the underlying anti-tumor mechanism of pPeOp deserves further research.…”

“…Due to the urgent clinical need, researchers are working to develop various antitumour drugs targeting gastric cancer. Omphalia lapidescens , a traditional Chinese medicine, has been reported to have a significant inhibitory effect on malignant tumors [1] . We previously identified a bioactive protein (pPe O p) from O .…”
