Pregnancy termination in dogs with non‐hormonal compounds: evaluation of selected derivatives

Abstract: Four non‐hormonal, pregnancy‐terminating compounds have been studied in bitches. One of these, 2‐(4‐chlorophenyl)‐s‐triazole[5,1‐a]isoquinoline (DL 717‐IT or L 12717), has been found to be very efficacious after a single subcutaneous injection. This compound appears to be promising for use in veterinary practice.

“…Our studies of conceptus elimination indicate that pregnancy arrest induced by DL 7 17-IT is not accompanied by uterine disturbances. This agrees with what was repeatedly demonstrated for DL 7 17-IT analogues by direct (uterine histology) or indirect (maintenance of fertility) means (Galliani & Lerner, 1976;Galliani et a/., 1981. For D L 717-IT, the above assertion is reinforced by the fact that no impairment of later reproductive performance was seen (Galliani.…”

“…For several years, we have been studying a new class of non-hormonal pregnancy-terminating compounds that arrest pregnancy in all the animal species tested. including the bitch (Lerner et al, 1975;Galliani & Lerner. 1976;Galliani ef al., 1980, 198 1).…”

“…2-Phenyl-triazole[S, 1 -alisoindoles and the corresponding isoquinolines are a unique class of non-hormonal anti-fertility agents with post-implantation activity. When administered subcutaneously or intramuscularly in single and multiple doses, these compounds terminate pregnancy in several species, including the bitch (Galliani & Lerner, 1976). Their activity was found to be dependent on the administered dose and the time of pregnancy.…”

“…Their activity was found to be dependent on the administered dose and the time of pregnancy. They are maximally effective when given immediately after implantation of fertilized ova, that is on day 4 of gestation in the hamster, day 7 in the rat and day 20 in the bitch (Galliani & Lerner, 1976, 1976aGalliani et al, 1977Galliani et al, , 1980Galliani et al, , 1981.…”
