Background: Phosphodiesterases (PDEs) play a major role in the regulation of cyclic adenosine monophosphate (cAMP)- and cyclic guanosine monophosphate (cGMP)-mediated pathways. Their inhibitors exhibit anti-inflammatory, vasodilatory and antithrombotic effects. Therefore, consumption of foods with PDE-inhibiting potential may possess beneficial influence on the risk of cardiovascular diseases. Methods: Four plant extracts (Arbutus unedo, Camellia sinensis, Cynara scolymus, Zingiber officinale) with promising ingredient profiles and physiological effects were tested for their ability to inhibit cAMP-specific PDE in vitro in a radioactive assay. Results: Strawberry tree fruit (Arbutus unedo) and tea (Camellia sinensis) extracts did not inhibit PDE markedly. Alternatively, artichoke (Cynara scolymus) extract had a significant inhibitory influence on PDE activity (IC50 = 0.9 ± 0.1 mg/mL) as well as its flavone luteolin (IC50 = 41 ± 10 μM) and 3,4-dicaffeoylquinic acid (IC50 > 1.0 mM). Additionally, the ginger (Zingiber officinale) extract and one of its constituents, [6]-gingerol, significantly inhibited PDE (IC50 = 1.7 ± 0.2 mg/mL and IC50 > 1.7 mM, respectively). Crude fractionation of ginger extract showed that substances responsible for PDE inhibition were in the lipoid fraction (IC50 = 455 ± 19 μg/mL). Conclusions: A PDE-inhibitory effect was shown for artichoke and ginger extract. Whether PDE inhibition in vivo can be achieved through ingestion of artichoke or ginger extracts leading to physiological effects concerning cardiovascular health should be addressed in future research.

This paper focuses on the effects of three thermal drying methods (microwave-, oven-and sun-drying) and one nonthermal drying method (freeze-drying) on the AOP of leaves of Vitex negundo and Vitex trifolia, which are consumed traditionally as herbal tea. Microwave-drying and freeze-drying were found to be able to maintain the AOP of the leaves but oven-drying and sun-drying resulted in deterioration of AOP. Microwave-drying has the advantage of short drying time and low water activity. AOP of Vitex herbal tea leaves that are microwave-dried and freeze-dried were not affected by storage up to 30 days. PRACTICAL APPLICATIONSTea, one of the most widely consumed beverages apart from water, has long been known for its health-promoting benefits in terms of its antioxidant properties due to its high phenolic content. Much of such studies, however, focus on green tea (Camellia sinensis). Although V. negundo and V. trifolia have been consumed traditionally as herbal tea, understanding of its antioxidant properties remains scarce. Drying serves as a vital part of tea processing, affecting its antioxidant content and appearance which in turn affects the commercial value of the tea. This study therefore serves as an important work in providing insights into the antioxidant properties of Vitex species and the best drying method of its leaves as herbal tea for commercial purpose. In addition, this study also provides insights into the effect of different drying methods on the storage of the leaves which is of value to the tea processing industry.

Four medicinal plants, Acorus calamus Linn., Alpinia galanga Willd., Desmodium triquetrum D.C. and Vitis discolour Dalz., were investigated for potential to inhibit cyclic AMP phosphodiesterase. The alcohol extract of Desmodium triquetrum was found to be most active. The chloroform and alcohol extracts of Desmodium triquetrum and Acorus calamus, and the petroleum ether and chloroform extracts of Alpinia galanga contain flavonoids. These are possibly the active principles responsible for their anti-bacterial activity.
