Preparation of Water Soluble Yeast Glucan by Four Kinds of Solubilizing Processes

Abstract: (1→3)-β-D-glucan from the inner cell wall of Saccharomyces cerevisiae is considered a member of a class of drugs known as biological response modifiers (BRM). However the glucan was an insoluble polysaccharide, which could be the major barrier to the utilization of glucan. In this case, the insoluble glucan was convent into a soluble form by four kind of solubilizing processes. The yield, solubility, chemistry structure and immunoprophylaxis efficacy of the soluble products were compared. Our date suggest that… Show more

“…4)) was seen to be similar to that of native glucan reported elsewhere (Du et al , 2012[9]; Saito et al , 1987[30]). The chemical shifts from GP were observed at 105.62, 87.19, 78.74, 76.47, 70.17, and 63.07 ppm, which correspond to the C-1, C-3, C-5, C-2, C-4, and C-6 respectively.…”

“…Thus, the NMR spectrum of the formulation verified the (1→3) β-D-glucan structure of GP. In addition, another small signal (C6 * ) was observed at 69.3 ppm, which is a characteristic peak due to β-1,6 branching (Du et al , 2012[ 9 ]). The degree of branching (DB) of the water-insoluble yeast glucan is reported to be as low as 0.003 (Kim et al , 2000[ 25 ]).…”

“…Particularly, the chemical ships observed at 4.48, 3.43, 3.59, 3.45, 3.47, and 3.82 ppm (Figure 4a) were assigned for H-1, H-2, H-3, H-4, H-5, and H-6, respectively, while chemical shifts corresponding to C-1, C-2, C-3, C-4, C-5, and C-6 (Figure 4a) appeared at 102.58, 73.32, 84.25, 68.16, 75.66, and 60.75 ppm, respectively. The assignment of all signals is in accordance with published spectra [21,39,45,46].…”

“…However, because of its insolubility, β3-glucan could not be used straightly for in vitro assays. Therefore, β3-glucan from CNCM I-3856 was phosphorylated in order to solubilize it prior to assay (Du, Zhang, Wang, & Xiao, 2012;Shi, Shi, & Li, 2014;Williams et al, 1991). Presence of phosphorylated glucan was first confirmed by observing a strong signal at 0 ppm on 1D 31 P-NMR (Supplementary Figure 4A).…”
