Presence and role of Simian Virus 40 (SV40) in malignant pleural mesothelioma

Abstract: Background. Evidence of a possible role of viruses in cancer first emerged in the early 1900s and was confirmed after the discovery of Epstein-Barr virus (EBV) in Conclusions. Simian virus 40 (SV40) is a small DNA virus from the genus polyomavirus, closely related to human polyomaviruses John Cunningham virus (JCV) and BK virus (BKV) and is highly oncogenic for rodents. The virus accidentally entered the human population through contaminated early batches of polio vaccine in the 1960s. After the discovery of S… Show more

“…Cases of mesothelioma without prior exposure to asbestos are rare and usually occur as a consequence of repeated mesothelial scarring or irradiation (Churg et al 2004). A role of Simian virus 40 in MPM pathogenesis has been speculated in the past, but consensus has not been achieved (Hmeljak and Cor 2009). Twenty-nine new MPM cases were diagnosed in Slovenia in 2007, accounting for an approximate incidence of 1.5 per 100,000.…”

“…The genome of SV40 is a single, circular, doublestranded DNA molecule functionally divided into three distinct regions: early, late and regulatory. The early region is expressed soon after entry into the cell, whereas the late region, encoding for capsid proteins, is expressed only after successful viral DNA replication within the permissive host cell 10 . Viral oncogenicity is mediated by the products of two genes encoded on the early region, large T (Tag) and small t (tag) antigen.…”
