Protective effect of resveratrol against corticosterone-induced neurotoxicity in PC12 cells

Abstract: Objective Resveratrol(RES) is a natural polyphenol which possesses an anti-depressant effect. However, the mechanisms of its anti-depressant effect remain unclear. The aim of the study is to investigate the potential mechanisms in the neuro-protective efficiency in the corticosterone-induced pheochromacytoma 12 (PC12) cells. Methods PC12 cells were treated with 200 μM of corticosterone in the absence or presence of different concentrations of RES for 24 h. Then, cell viability was measured by Cell Counting Kit… Show more

“…In the current study, the intracellular Ca 2+ concentration was increased, the protein expression of Bax was enhanced, and that of Bcl-2 was attenuated in PC12 cells treated with corticosterone. These results are consistent with those of other studies [8,26,46]. By contrast, we confirmed that these effects were reversed in PC12 cells pretreated with WEST.…”

“…We next assessed the expression of the mitochondrial apoptotic pathway-related proteins cytochrome c, caspase-9, and caspase-3. Corticosterone treatment upregulated cytochrome c, caspase-9, and caspase-3 protein expression, consistent with other studies [8,26,46,52]. By contrast, pretreatment with WEST downregulated the expression of these proteins.…”

“…Interestingly, Y. Zhang et al (2019) conducted an in vitro study to evaluate the protective effects of resveratrol against corticosterone‐induced neurotoxicity in pheochromocytoma 12 (PC12) cells. They discovered that various concentrations of resveratrol (2.5, 5, and 10 μM) enhanced cell viability and inhibited PC12 cell apoptosis by suppressing apoptosis‐related proteins, such as Bax and caspase‐3, and increasing Bcl‐2 protein expression.…”

“…The Chinese have utilized this plant for decades to treat a variety of inflammatory disorders. In recent studies, SLDS has strong protective benefits against hepatitis, colitis, skeletal muscle atrophy, and myocardial damage by reducing inflammation (Hu et al 2014, Huang et al 2019, Liu et al 2019. SLDS performs a neuroprotective role in preclinical models of Alzheimer's disease (AD) and cerebral ischemia by modulating microglial activation and distribution (Zhang, Wang, et al 2016.…”
