Protective Effects of Lactoferrin against SARS-CoV-2 Infection In Vitro

Abstract: SARS-CoV-2 is a newly emerging virus that currently lacks curative treatments. Lactoferrin (LF) is a naturally occurring non-toxic glycoprotein with broad-spectrum antiviral, immunomodulatory and anti-inflammatory effects. In this study, we assessed the potential of LF in the prevention of SARS-CoV-2 infection in vitro. Antiviral immune response gene expression was analyzed by qRT-PCR in uninfected Caco-2 intestinal epithelial cells treated with LF. An infection assay for SARS-CoV-2 was performed in Caco-2 cel… Show more

“…In fact, during Huh 7 cell treatment with LF, up-regulation of the expression of several interferon-stimulated genes (MX1, ISG15, IFITM3, and RSAD2 that encodes radical S-adenosyl methionine domain-containing protein 2 also known as Viperin) was observed (Mirabelli et al, 2020). Similarly, although the addition of LF (100 µg/mL) induced a partial inhibition of SARS-CoV-2 multiplication in pre-infected Caco-2 intestinal epithelial cells, this protein significantly induced and upregulated the expression of many innate and adaptive immunity markers, such as IFNA1, IFNB1, TLR3, TLR7, IRF3, IRF7 and MAVS (Salaris et al, 2021). Based on this in vitro potential of bLF against SARS-CoV-2, the authors suggested that LF combined with Vitamin D will be valid adjuvant therapeutic tool for patients with COVID-19 (Salaris et al, 2021).…”

“…In the last year, there has been accumulating evidence from studies in vitro that bLf is active against SARS-CoV-2 [ 22 , 34 , 45 ]. BLf exerts its antiviral activity either by obscuring the host cell receptors [ 22 ] or by directly binding to the SARS-CoV-2 spike glycoproteins [ 34 ].…”

“…It may also inhibit SARS-CoV-2 by binding to some of the receptors required for viral entry, such as ACE2 and HSPGs ( 42 , 49 ). An in vitro study confirmed that lactoferrin inhibits SARS-CoV-2 infection and replication in Caco-2 cells ( 50 ). In addition, an in vivo study confirmed that lactoferrin accelerates SARS-CoV-2 RNA-negative conversion in patients with COVID-19 ( 51 ).…”
