Pulsed-Field Gel Electrophoresis of Salmonella enterica

Abstract: Pulsed-field gel electrophoresis (PFGE) is considered a "gold standard" for the molecular characterization of a number of bacterial strains. Its strength relies on its high discriminatory power, together with its high reproducibility. For many years, an international network, PulseNet International, allows the rapid comparison of PFGE data obtained all over the world, and it provides a valuable tool to promptly recognize the epidemiological dynamics of many pathogens, including Salmonella enterica. Here we des… Show more

“…Pulsed-field gel electrophoresis (PFGE) of the isolates was carried out following the PulseNet One-day Standardized PFGE Protocol for Salmonella with minor modifications (Camarda et al, 2015). Restriction endonuclease digestion was carried out using XbaI (Takara, Dalian, China).…”

“…Subtyping methods must fulfil many requirements such as high discriminatory power, easy performance, clear interpretation and possibility for international standardization. Currently, pulsed-field gel electrophoresis (PFGE), which has been internationally standardized, is considered as the golden standard for genotyping of Salmonella and it is the only generic molecular method suitable for all Salmonella serotypes [ 14 16 ]. However, not all the isolates unrelated to an outbreak are different by PFGE from the outbreak-related isolates and PFGE has been shown to suffer from a high variation between the different laboratories.…”

“…The pulsed field gel electrophoresis (PFGE) of Salmonella is carried out in accordance with PulseNet’s 1-day standardized PFGE protocol, with slight modifications ( Camarda et al, 2015 ). Xba I (Takara, Dalian, China) was used for restriction endonuclease digestion.…”
