Qualitative analysis of Psoraleae Fructus by HPLC‐DAD/TOF‐MS fingerprint and quantitative analysis of multiple components by single marker

Abstract: A variety of bioactive substances may account for the recognized efficacy and wide clinical application of Psoraleae Fructus in China. A high-performance liquid chromatography-diode array detector (HPLC-DAD) fingerprint method was developed to present the comprehensive phytochemical profile of the crude drug. Thirteen major compounds were separated and identified by HPLC coupled with time-of-flight mass spectrometry (HPLC/TOF-MS), namely psoralenoside (PO), isopsoralenoside (IPO), psoralen (PS), isopsoralen (I… Show more

“…Compound 34 showed a protonated molecular ion at m/z 187.0393 at 10.77 min, and the MS/MS spectrum generated fragment ions, including m/z 159.0439 [M+H‐CO] + , 143.0489 [M+H‐CO 2 ] + , 131.04912 [M+H‐2CO] + and 115.0543 [M+H‐CO 2 ‐CO] + . By comparing the obtained mass spectra with the previous literature, compound 34 was tentatively assigned to be isopsoralen [22]. The proposed fragmentation schemes are shown in Figure 3B.…”

“…The other 23 compounds were characterized as isoflavones, chalcones, and flavonones. Ten of them (compounds 44 , 45 , 71 , 79 , 93 , 103 , 110 , 118 , 127 , and 133 ) were confirmed by comparing with standard compounds and the left constituents (compounds 25 , 29 , 35 , 68 , 103 , 108 , 117 , 98 , 131 , 33 , 52 , 127 , and 136 ) were tentatively identified by comparing with their accurate mass measurements of both MS spectra within the mass error ± 5 ppm and literature .…”

“…The HPLC four stage rod-electrostatic field orbit trap higher solution mass spectrometry analysis results of knocked-out components were shown in Figure 4 . Protonated ions, adducted ions, and fragment ions of each peak were then elucidated in accordance with reference HPLC chromatogram of standard solutions and related data in the literatures [ 35 , 36 , 37 , 38 , 39 , 40 , 41 ]. The formula of compound P2 with [M + H] + ions at m / z 254, C 15 H 10 O 4 , produced ion fragments of 136/118, and were tentatively characterized as daidzein ( Figure 5 a).…”
