Quantitative determination of atractylon in Atractylodis Rhizoma and Atractylodis Lanceae Rhizoma by 1H-NMR spectroscopy

Abstract: (1)H-NMR spectroscopy was successfully applied to the quantitative determination of atractylon in Atractylodis Rhizoma (dried rhizomes of Atractylodes ovata and A. japonica) and Atractylodis Lanceae Rhizoma (dried rhizomes of Atractylodes lancea and A. chinensis). The analysis was carried out by comparing the integral of the H-12 singlet signal of atractylon, which was well separated in the range of delta 6.95-7.05 ppm in the NMR spectrum, with the integral of a hexamethyldisilane (HMD) signal at delta 0 ppm. … Show more

“…18 The LOQ values typically vary from 3 to 20 ng/mL in LC-MS methods. 20,21,26 However, the levels of LOD and LOQ values found in this study are comparable with other published applications of NMR in the field of pharmaceutical research, 27,28 including those using a higher magnetic field. 29,30 Although LOD and LOQ values may be lower using other methods (i.e., the HPLC method), the focus of this study is to quantify the amount of TNF release in biological fluid simulant rather than determine its aqueous solubility limit.…”

“…The ethyl acetate fraction was concentrated under reduced pressure to yield each aglycone. The purity of each aglycone thus obtained was estimated by quantitative 1 H NMR analysis (24). Geniposide, genipin, and loganin were obtained from Wako Pure Chemicals (Osaka, Japan).…”

“…In a previous paper, we reported the quantitative determination of the components of crude drugs by proton-specific quantitative NMR ( 1 HqNMR) [12][13][14][15][16]. The 1 H-qNMR method has various advantages over other quantitative methods, including HPLC.…”
