Raising the NKT cell family

Abstract: Natural killer T cells (NKT cells) are CD1d-restricted, lipid antigen-reactive, immunoregulatory T lymphocytes that can promote cell-mediated immunity to tumors and infectious organisms, including bacteria and viruses, yet paradoxically they can also suppress the cell-mediated immunity associated with autoimmune disease and allograft rejection. Furthermore, in some diseases, such as atherosclerosis and allergy, NKT cell activity can be deleterious to the host. Although the precise means by which these cells ca… Show more

“…Once committed to the NKT cell lineage, numerous transcription factors, cytokines, chemokines, and costimulatory molecules have been documented that are uniquely required for maturation and homeostasis of NKT cells, but dispensable for conventional T cell development. At least two key control points exist in NKT cell development, which govern NKT cell selection to give rise to immature NKT cells branched from the conventional T cell development pathway and NKT cell maturation with a series of phenotypic and functional changes, respectively [3,21] . The basic pathway of NKT cell development is illustrated in Figure 1.…”

“…Unlike conventional T cells, which respond to foreign peptides presented by MHC class І or ІІ molecules through interaction with their αβ TCR, NKT cells recognize antigenic glycolipids presented by the MHC class І-like molecule CD1d [3,4] . Mice with deletion of CD1d gene eradicates NKT cells including both type І and type ІІ.…”

“…-2136 Introduction iNKT cells express a panoply of NK-cell receptors [1] and a canonical TCR through which they recognize (glyco-)lipid antigens [2]. iNKT cells activate similar signaling cascades after TCR ligation like other T lymphocytes [3], but utilize unique transcription factors for their development such as the promyelocytic leukemia zinc finger (PLZF) [4][5][6]. According to two different developmental models PLZF characterizes distinct maturation stages and polarized subsets.…”
