RANKL expressions in preservation of surgical tooh extraction treated with Moringa (Moringa oleifera) leaf extract and demineralized freeze-dried bovine bone xenograft

Abstract: Background: Preservation of sockets is a procedure aimed to reduce bone resorption after tooth extraction. One of the most commonly used xenograft materials is demineralized freeze-dried bone bovine xenograft (DFDBBX). Meanwhile, one of the key regulations in osteoclast genesis process is RANKL bond. A decrease in the number of RANKL expressions can suppress the osteoclast genesis process so that bone resorption can be prevented. The combination of Moringa leaf extract and DFDBBX, as a result, is expected to d… Show more

“…Most experiments confirmed ethanol extract upper hand. 6,7,17,18,[22][23][24][25][26][27][28] While, Kwon research confirmed superior results for water extract of moringa over ethanolic extract (Table 2). 16 Previous studies confirmed the tissue preservation of M. oleifera leaf ethanolic extracts, for example it protected rats against the hepatotoxicity of various drugs, 9 and increased cell proliferation and cell migration of human dermal fibroblasts.…”

“…Going into some details, the MO leaf extract increased alkaline phosphatase activity, collagen type I expression, osteocalcin and transforming growth factor beta (TGF B ) expressions (responsible for deposition of bone matrix ), besides it decreased number of Rankle expression . 1,24,29,31,32 Moreover, a study conducted on upper central extraction socket in a rat model 2018 using mate tea intra-gastric lavage concluded positive effects on alveolar healing at 28 days postoperatively. 4 Our study uniquely used high concentration of water extract (11.7%) suggested to be comparable to ethanol osteoprotective property.…”

“…Tooth extraction is one of the most frequently performed procedures in dental practice. Tooth extraction is carried out because of terminal condition or unresponsive response towards current treatment, such as severe caries, periodontal disease, impaction, fracture, teeth affected by cysts or tumors [1]. Traumatic tooth extraction leads to unfavorable healing process, resulting excessive bone resorption that will interfere future denture treatment [2].…”

“…DFDBBX will function as a scaffold or framework in the growth of new bone originating from the formation of osteoblasts of the socket. Whereas the anti-inflammatory properties in Moringa leaves will suppress the inflammatory response in the socket after tooth extraction [1]. Therefore, this study was aimed to determine the induction effect of the combination of Moringa oleifera and DFDBBX extracts on the growth of woven bone of the alveolar bone socket.…”

“…After that, TNF receptor-associated factor 6 (TRAF6), NF-B, c-Jun N-terminal kinase (JNK)/cJun/fos, and nuclear factor of activated T cells (NFAT) begin to differentiate precursor osteoclasts into preosteclast cells. About 1-4 weeks needed for resorption to take place by osteoclast mediation process (3) .…”
