Rapid, label-free, and sensitive point-of-care testing of anti-SARS-CoV-2 IgM/IgG using all-fiber Fresnel reflection microfluidic biosensor

Abstract: The ongoing global pandemic of SARS-CoV-2 has promoted to develop novel serological testing technologies since they can be effectively complementary to RT-PCR. Here, a new all-fiber Fresnel reflection microfluidic biosensor (FRMB) was constructed through combining all-fiber optical system, microfluidic chip, and multimode fiber bio-probe. The transmission of the incident light and the collection and transmission of Fresnel reflection light are achieved using a single-multi-mode fiber optic coupler (SMFC) witho… Show more

“…The recognition elements of optical biosensors can be divided into aptamers, molecular imprint polymers (MIPs), and antibodies [ 49 ]. Wenjuan and his colleagues created the first unique microfluidic biosensor using Fresnel reflection for the detection of SARS-CoV-2 without a label that is quick, simple, and sensitive [ 50 ].…”

“…They used a photodiode detector (PD-1000) that can quantify antibody concentrations by measuring the intensity of the Fresnel reflection light that changes along with the increase or decrease in the refractive index (RI) of the biosensing surface due to the formation of antigen-antibody complexes. This device can detect anti-S IgG/IgM within 7 min with a limit of detection of 0.82 ng/mL for IgG and 0.45 ng/mL for IgM, which is validated by testing six negative serum samples ( Table 1 ) [ 32 ].…”

“…The specific IgG was proved with a detection limit of 12.5 ng/mL for a sample sized 100 µL and an assay lasting 25 min. Xu et al constructed another fiber optic device for the detection of IgG and IgM against S protein [ 90 ]. The researchers opted for a Fresnel reflection microfluidic biosensor as a device allowing quite fast and label-free detection of the antibodies.…”
