Derivatization of two monophosphorylated positional isomers of insulin receptor peptide and triply phosphorylated insulin receptor peptide, all with the same amino acid sequence, with 9‐fluorenylmethyl reagents (chloroformate, Fmoc‐Cl, or N‐succinimidyl carbonate, Fmoc‐OSu), allowed efficient separation of phosphorylated peptides from nonphosphorylated peptides by microchip CE using 30 mM ammonium acetate BGE in mixed hydro‐organic 40/40 v/v water/methanol solvent and and on‐chip ESI/MS detection . The derivatization enabled not only the separation of monophosphorylated positional peptide isomers but also identification of the phosphorylation site by tandem MS detection.…”
